采用单因素考察和响应面分析法相结合的方法,对提取液pH、渗漉流速、料液比3个影响总生物碱提取率的关键因素进行优化,建立了苦参中总生物碱的提取模型,回归模型显著。最终确定提取优化条件为提取液pH=2.76,以3.22mL/(kg·min)的渗漉流速,收集4.31倍量的渗漉液。在此最佳提取条件下,通过验证实验得出苦参总生物碱提取率为94.5%,与预测值94.932%较为接近。试验得到结论,Box-Bohnken设计结合响应面分析法可很好地对苦参总生物碱提取工艺进行优化。
The combination of single factor analysis and response surface analysis was used to optimize the key factors that affect the total alkaloid extraction rate, such as the pH of the extract, the flow rate of percolation and the ratio of solid to liquid. The extraction model of total alkaloids from Sophora flavescens The regression model is significant. Finally, the optimal extraction conditions were determined as pH = 2.76 and the percolation rate of 3.31mL was collected with the percolation rate of 3.22mL / (kg · min). Under the optimal extraction conditions, the yield of total alkaloids from Sophora flavescens L. was 94.5%, which was close to the prediction value of 94.932%. The conclusion of the experiment is that Box-Bohnken design combined with response surface methodology can be used to optimize the total alkaloid extraction process of Sophora flavescens.
