目的:研究离体人肠道菌群对头花蓼提取物中槲皮素、槲皮苷和金丝桃苷3个单体成分在体外代谢转化的情况。方法:将3个成分与人肠道菌孵育液分别在37℃离体厌氧环境中培养24 h,采用超高压液相色谱-电喷雾-四级杆-飞行时间串联质谱仪进行检测,运用Metabolite Predict、Metabolite Detect对样品中的代谢产物进行预测并检测分析,结合Mass Defect Filter技术进一步有效提取代谢产物信息,排除基质背景干扰,并应用Data Analysis中Smart Formula功能推测代谢产物化学式。结果:槲皮苷和金丝桃苷主要以脱糖反应为主,苷元槲皮素进一步发生羟基化、甲基化、开环、还原等反应。结论:该实验建立了人肠道厌氧菌群体外代谢研究模型,为头花蓼提取物在人肠道内转化吸收提供了科学依据。
OBJECTIVE: To study the metabolic transformation of three monomer components of quercetin, quercitrin and hyperoside in Polygonum capitatum L. extract isolated from human intestinal flora in vitro. Methods: The three components were incubated with human intestinal bacteria in anaerobic environment at 37 ℃ for 24 h, respectively. The concentrations of the three components were determined by using ultra-high pressure liquid chromatography-electrospray-quadrupole-time of flight tandem mass spectrometry Metabolite Predict, Metabolite Detect The metabolites in the sample are predicted and detected. Mass Defect Filter technology is used to further extract the metabolite information to eliminate the matrix background interference. Results: Quercitrin and hyperoside mainly de-saccharification reaction, aglycone quercetin further hydroxylation, methylation, ring opening, reduction and other reactions. Conclusion: This experiment established an in vitro model of human intestinal anaerobic bacterial metabolism, which provided a scientific basis for the transformation and absorption of Polygonum capitatum extract in human intestine.
