通过响应面法确定不同年份黄精中总糖含量的最佳测定条件,在单因素试验的基础上,选定浓硫酸质量分数、蒽酮用量和显色时间3个因素,利用响应面分析法考察各因素对吸光度的影响,结合3,5-二硝基水杨酸法对黄精中还原糖含量进行测定,并比较不同年份中多糖含量。结果表明:最优测定条件为蒽酮用量0.2 g,浓硫酸质量分数为87%,显色时间为25 min;经过测定,4年生黄精总糖和还原糖含量均高。该方法适合黄精总糖含量的测定,可用于评价黄精药材的质量和为黄精的采收时间提供理论依据。
Response surface method to determine the optimum conditions for determination of total sugar content of Polygonatum sibiricum in different years, on the basis of single factor test, the selected concentration of concentrated sulfuric acid, anthrone dosage and color development time of three factors, using response surface methodology The effect of various factors on the absorbance, combined with 3,5-dinitrosalicylic acid method for determination of reducing sugar content in Polygonatum, and compare the polysaccharide content in different years. The results showed that the optimum conditions were as follows: anthrone dosage 0.2 g, concentrated sulfuric acid 87%, and colorimetric time 25 min. The contents of total sugar and reducing sugar of 4-year-old Polygonatum were all high. The method is suitable for the determination of the total sugar content of Polygonatum sibiricum and can be used for evaluating the quality of Polygonatum sibiricum and providing the theoretical basis for the recovery time of Polygonatum sibiricum.
