目的采用响应面法优化超临界流体技术制备姜黄素脂质体的处方工艺。方法运用中心复合设计考察磷脂质量浓度、脂药质量比和pH值三因素对姜黄素脂质体包封率的影响及其交互作用。根据实验数据建立二次回归模型,并进行响应面分析。结果超临界流体技术制备姜黄素脂质体的工艺可行,且最佳处方配比为:磷脂质量浓度51.16 g·L-1、脂药质量比24.04∶1、pH值6.28。结论该条件下姜黄素脂质体包封率为80.43%,与模型预测值79.55%偏差较小,说明预测模型可靠性较高,可用于处方工艺的优化。
Objective To optimize the formulation of curcumin liposomes by using supercritical fluid technology with response surface methodology. Methods The center complex design was used to investigate the effect of three factors on the entrapment efficiency of curcumin liposomes and the interaction between them. Based on the experimental data to establish a quadratic regression model, and response surface analysis. Results The technology of supercritical fluid technology for preparation of curcumin liposomes was feasible. The optimal prescription ratio was 51.16 g · L-1, the mass ratio of lipid to drug was 24.04:1, pH value was 6.28. Conclusions The encapsulation efficiency of curcumin liposomes is 80.43% under the conditions, which is less than the predicted value of 79.55%. This indicates that the prediction model is more reliable and can be used to optimize the formulation process.
