目的 检测妊娠高血压综合征 (妊高征 )胎盘滋养细胞浸润能力 ,胰岛素样生长因子 II(IGF II)、转化生长因子 β1(TGF β1)对细胞滋养细胞侵袭力的影响。 方法 从正常及妊高征胎盘分离、纯化细胞滋养细胞行体外培养 ;用体外侵袭试验检测正常及妊高征胎盘滋养细胞的侵袭力 ;IGF II、TGF β1对两组滋养细胞侵袭力的影响。 结果 妊高征滋养细胞较正常滋养细胞的侵袭力显著降低 (P <0 .0 5 ) ;IGF II明显促进、TGF β1明显抑制正常妊娠滋养细胞的侵袭能力(P <0 .0 5 ) ;二者对妊高征滋养细胞侵袭力的影响不显著 (P >0 .0 5 )。 结论 妊高征胎盘滋养细胞的侵袭力显著降低 ;IGF II、TGF β1可能在妊高征胎盘滋养细胞浅浸润过程中起一定作用。
Objective To investigate the effect of placental trophoblast invasion, insulin-like growth factor II (IGF II) and transforming growth factor-β1 (TGF-β1) on the invasiveness of cytotrophoblasts in pregnancy induced hypertension syndrome (PIH). Methods The placental cells from normal and pregnancy induced hypertension were cultured and purified in vitro. The invasiveness of placental trophoblast cells in normal and pregnancy induced hypertensive rats was detected by in vitro invasion assay. The effects of IGF II and TGF β 1 on the invasiveness of trophoblast cells in both groups were analyzed. Results The invasiveness of pregnancy-induced trophoblastic cells was significantly lower than that of normal trophoblast cells (P <0.05), while IGF II was significantly increased. TGF β 1 significantly inhibited the invasive ability of normal gestational trophoblast cells (P <0.05) The effect of PIH on the invasiveness of PIH was not significant (P> 0.05). Conclusions The invasiveness of placental trophoblast cells in pregnancy induced hypertension significantly decreases. IGF II and TGFβ1 may play a role in the induction of placental trophoblast cells with shallow infiltration.
