目的改善肿瘤细胞B7分子表达。方法将小鼠ALA9702肺癌细胞与活化的B淋巴细胞用PEG进行细胞融合,经ELISA及免疫细胞化学筛选后,观察了融合细胞的体外生长曲线,并用S-P免疫细胞化学染色观察了融合细胞的肺癌抗原及B7分子的表达。结果经筛选获得1株融合细胞FLB2C,既表达肺癌抗原又表达B7分子,该细胞在体外培养中贴壁性减弱,生长变慢。结论通过细胞融合方法可以有效改善肿瘤细胞的B7分子表达,从而提高肿瘤细胞的抗原性,为制备更有效的肿瘤疫苗奠定了基础
Objective To improve the expression of B7 in tumor cells. Methods The murine ALA9702 lung cancer cells and activated B lymphocytes were fused with PEG. The growth curve of the fused cells was observed by ELISA and immunocytochemistry. The lung cancer antigens of the fused cells were observed by SP immunocytochemistry And B7 molecule expression. Results One fusion cell, FLB2C, was obtained by screening, which expressed both lung cancer antigen and B7 molecule. The cell adhesion decreased and grew slowly in vitro. Conclusion The cell fusion method can effectively improve the expression of B7 in tumor cells and enhance the antigenicity of tumor cells, which lays the foundation for the preparation of more effective tumor vaccines
