目的测定慢性特发性血小板减少性紫癜(CITP)小鼠外周血中IL-11及其受体(IL-11R)的水平,探讨CITP的发病机制,为ITP的治疗提供新的理论依据。方法BALA/C小鼠60只,随机分为对照组(20只)和CITP组(40只),通过腹腔注射豚鼠抗小鼠血小板血清(APS)的方法建立CITP模型。于造模第8d分别采用ELISA、流式细胞术测定两组小鼠外周血中IL-11及IL-11R的水平。结果CITP小鼠外周血中IL-11及IL-11R平均水平均较对照组升高(P<0.05);其中有31只小鼠同时存在IL-11及IL-11R水平的升高,余9只小鼠仅存在IL-11水平的升高,而IL-11R水平降低。结论IL-11及IL-11R在CITP的发病机制中起一定作用,但它们发挥作用的原理及过程尚需进一步探讨。
Objective To determine the levels of IL-11 and its receptor (IL-11R) in peripheral blood of chronic idiopathic thrombocytopenic purpura (CITP) mice and to explore the pathogenesis of CITP, providing a new theoretical basis for the treatment of ITP. Methods Sixty BALB / C mice were randomly divided into control group (n = 20) and CITP group (n = 40). CITP model was established by intraperitoneal injection of guinea pig anti-mouse platelet serum (APS) On the 8th day after modeling, the levels of IL-11 and IL-11R in the peripheral blood of the two groups were measured by ELISA and flow cytometry respectively. Results The average levels of IL-11 and IL-11R in peripheral blood of CITP mice were significantly higher than those of the control group (P <0.05). Eleven of the mice had elevated levels of IL-11 and IL-11R at the same time Only mice had elevated levels of IL-11 only, whereas levels of IL-11R decreased. Conclusion IL-11 and IL-11R play a role in the pathogenesis of CITP, but the principle and process of their role need to be further explored.
