目的 研制识别癌胚抗原的基因工程单链抗体,使其能够应用于肿瘤的免疫诊断和治疗。方法 利用RTPCR 方法扩增抗体可变区基因并表达于噬菌体表面。通过免疫筛选获得癌胚抗原特异单链抗体CL3scFv 。结果 ELISA 和免疫组化均显示抗体CL3scFv 能够结合CEA,而不与正常胃肠道组织起反应。结论 该抗体将成为胃肠道恶性肿瘤辅助诊断和治疗的有效试剂。
Objective To develop a genetically engineered single-chain antibody recognizing carcinoembryonic antigen that can be used for the immunodiagnosis and treatment of tumors. Methods RT-PCR was used to amplify the antibody variable region and express it on the phage surface. Through immunoscreening, a single-chain antibody, SC-3, scFv was obtained. Results Both ELISA and immunohistochemistry showed that the antibody CL 3 -scFv was able to bind to CEA but did not react with normal gastrointestinal tissue. Conclusion This antibody will be an effective reagent for the diagnosis and treatment of gastrointestinal malignancy.
