作者用设计合成的两对引物建立了巢式PCR-EB染色法检测B19DNA新技术,扩增产物为104bp片段,敏感性达0.007fg,是一般PCR的100倍,是一种特异、敏感、简便、快速诊断B19感染的新方法.用该方法检测了57例自然流产和死胎的胎儿组织,17例阳性,其中自然流产11/42(26%),死胎6/15(40%),证实了国内胎儿有B19病毒感染,和自然流产、死胎可能相关,并对PCR实验中防污染措施进行了探讨.
The authors designed a nested PCR-EB method to detect B19 DNA by using two pairs of primers designed and synthesized. The amplified product was 104bp with a sensitivity of 0.007fg, which was 100 times of that of general PCR. It was a specific, sensitive, A Simple and Rapid Method to Diagnose B19 Infection. Fifty-seven fetuses with spontaneous abortion and stillbirth were detected by this method, of which 17 were positive, including 11/42 (26%) spontaneous abortion and 6/15 (40%) stillbirth, confirming the presence of B19 virus infection in domestic fetuses and natural Abortion and stillbirth may be related, and the anti-pollution measures in PCR experiments were discussed.
