目的:探讨垂体肿瘤转化基因(pituitary tumor-transforming gene,PTTG)在子宫内膜癌的蛋白表达水平及其临床意义。方法:采用免疫组化技术对子宫内膜癌患者、增生性子宫内膜及正常健康者的子宫内膜组织中的PTTG的表达情况进行测定,通过结合临床病理特征,初步研究PTTG基因在子宫内膜癌的发生与发展中的作用及其临床应用价值。结果:子宫内膜癌组织中的PTTG的表达阳性率明显高于正常子宫内膜组织和增生性子宫内膜组织(P<0.05)。手术-病理分期为Ⅲ~Ⅳ期患者的PTTG蛋白表达阳性率明显高于Ⅰ~Ⅱ期,即随着病理分期的增加PTTG阳性率增高(χ2=5.22,P<0.05);高分化腺癌(G1)的PTTG蛋白表达阳性率明显低于中分化和低分化腺癌(G2和G3)(χ2=4.73,P<0.05);发生淋巴结转移、肌层浸润超过1/2的患者PTTG蛋白表达阳性率明显高于未发生淋巴结转移、肌层浸润≤1/2的患者(χ2=4.25、6.31,P<0.05)。结论:PTTG蛋白的过表达参与了子宫内膜癌发生和发展,与子宫内膜癌的恶性程度密切相关,检测患者子宫内膜组织的PTTG蛋白表达可用来鉴别肿瘤恶性程度及预测预后。
Objective: To investigate the protein expression of pituitary tumor-transforming gene (PTTG) in endometrial carcinoma and its clinical significance. Methods: The expression of PTTG in endometrial cancer patients, proliferative endometrium and normal healthy endometrial tissues was detected by immunohistochemical technique. PTTG gene was preliminarily studied in the uterus by combining with clinicopathological features The occurrence and development of membranous cancer and its clinical value. Results: The positive rate of PTTG expression in endometrial carcinoma tissues was significantly higher than that in normal endometrial tissues and proliferative endometrial tissues (P <0.05). The positive rate of PTTG expression in patients with stage Ⅲ-Ⅳ disease was significantly higher than that in stage Ⅰ-Ⅱ, that is, the positive rate of PTTG was increased with the increase of pathological stage (χ2 = 5.22, P <0.05); well-differentiated adenocarcinoma G1) was significantly lower than that of moderately differentiated and poorly differentiated adenocarcinoma (χ2 = 4.73, P <0.05). The positive expression of PTTG protein in patients with lymph node metastasis and myometrial invasion more than 1/2 The rates were significantly higher than those without lymph node metastasis and myometrial invasion ≤ 1/2 (χ2 = 4.25, 6.31, P <0.05). Conclusion: Overexpression of PTTG protein is involved in the occurrence and development of endometrial carcinoma, which is closely related to the malignancy of endometrial carcinoma. Detecting PTTG protein expression in endometrial tissue can be used to identify the degree of malignancy and prognosis.
