[目的]探讨大豆的皂甙(Soyasaponin,SS)对实验性大鼠矽肺纤维化模型中肺组织细胞Fas/FasL表达的影响。[方法]通过复制大鼠矽肺纤维化模型,观察各组大鼠血清中超氧化物歧化酶(SOD)、肺组织丙二醛(MDA)在染尘及SS干预后的含量变化。采用免疫组化法观察各组大鼠肺组织细胞Fas和FasL表达的改变。[结果]给予SS后,与对模型组相比血清SOD升高、MDA显著降低(P﹤0.05)。SiO2组Fas蛋白表达为相应对照组的4.51和5.05倍,其阳性信号主要集中于支气管上皮细胞、肺泡上皮细胞。[结论]Fas/FasL系统可能参与矽肺纤维化的发病过程,一定剂量的大豆皂甙可能通过降低氧化应激,从而缓解矽肺纤维化的进展。
[Objective] To investigate the effect of Soyasaponin (SS) on the expression of Fas / FasL in the lung tissue of experimental silicotic fibrosis model in rats. [Methods] The changes of serum superoxide dismutase (SOD), malondialdehyde (MDA) content in lung tissue and the content of SS after intervention were observed by duplicating rat model of silicotic fibrosis. Immunohistochemistry was used to observe the changes of Fas and FasL expression in lung tissue of rats in each group. [Results] Compared with model group, serum SOD increased and MDA decreased significantly (P <0.05). The expression of Fas protein in SiO2 group was 4.51 and 5.05 times higher than that in the corresponding control group, and the positive signals of SiO2 were mainly in bronchial epithelial cells and alveolar epithelial cells. [Conclusion] The Fas / FasL system may participate in the pathogenesis of silicosis. A certain dosage of soybean saponin may alleviate the progress of silicosis by reducing oxidative stress.
