本文采用反相高效液相色谱法,检测了用苯异硫腈柱前衍生的氨基酸;对小麦在水分胁迫条件下的12种有关氨基酸进行了实际分析的应用。方法上不采用通常的浓度梯度洗脱,而采用流速梯度洗脱;除组氨酸与酪氨酸合为一峰不能分离外,其余均能良好分离,且重复性好,保留时间最大偏差仅为0.152分,峰百分面积最大偏差仅为0.60%。检测结果能反映出小麦叶片氨基酸代谢的变化情况与抗旱性之间有一定联系。
In this paper, reversed-phase high performance liquid chromatography (HPLC) was used to detect amino acids derived from pre-column with benzene isothiocyanide. The practical application of 12 amino acids under water stress was analyzed. The method does not use the usual concentration gradient elution, but the use of gradient flow elution; except histidine and tyrosine as a peak can not be separated, the rest are well separated, and good repeatability, the maximum retention time deviation of only 0.152 points, the maximum percentage deviation of the peak area is only 0.60%. The test results can reflect the change of amino acid metabolism in wheat leaves and drought resistance have a certain relationship.
