外周血干细胞和骨髓基质细胞联合输注对急性放射病造血重建作用的实验研究

目的 观察外周血干细胞联合骨髓基质细胞输注对急性放射病造血功能恢复的影响。方法  8 5Gyγ射线照射的BALB c小鼠 ,分别输入红细胞 (对照组 )、动员的外周血干细胞 (PB SCT组 )、外周血干细胞联合扩增的骨髓基质细胞 (实验组 ) ,观察受体鼠 4周的生存率、骨髓有核细胞 (BMNC)、粒细胞巨噬细胞集落形成单位 (CFU GM)、成纤维细胞集落形成单位 (CFU F)、外周血白细胞计数 (WBC)等指标。结果 实验组的生存率、BMNC、CFU GM、CFU F较对照组和PBSCT组显著升高 ,P <0 0 5 ;WBC的最低值较其他两组高 (P <0 0 1) ,WBC恢复较PBSCT组快 (P <0 0 1)。结论外周血干细胞联合骨髓基质细胞输注有促进急性放射病造血重建的作用 ,其作用优于单纯的外周血干细胞输注 (移植 )。 Objective To observe the effects of peripheral blood stem cells combined with bone marrow stromal cells infusion on hematopoietic function recovery after acute radiation sickness. Methods 8 BALB / c mice irradiated with 5 Gy γ-rays were injected with erythrocytes (control group), mobilized peripheral blood stem cells (PB SCT group), peripheral blood stem cells and expanded bone marrow stromal cells (experimental group) Week survival rate, BMNC, CFU GM, CFU F, WBC and so on. Results The survival rate, BMNC, CFU GM and CFU F in the experimental group were significantly higher than those in the control group and PBSCT group (P <0.05). The lowest WBC in the experimental group was higher than the other two groups (P <0.01) The PBSCT group was faster (P <0.01). Conclusion Peripheral blood stem cells combined with bone marrow stromal cells can promote the hematopoietic reconstitution of acute radiation sickness, and its effect is better than that of pure peripheral blood stem cell transplantation.
