目的观察白桦脂醇和地塞米松共同作用于小鼠MC3T3-E1细胞后,内源性脂质蓄积与细胞凋亡中的关系。方法以MC3T3-E1细胞为研究对象,采用白桦脂醇和地塞米松共同作用于MC3T3-E1细胞,测定细胞内甘油三酯含量;NileRed荧光染色观察脂质蓄积情况;流式细胞技术测定成骨细胞凋亡率;ELISA法检测Caspase-3酶活性;采用Spearman等级相关进行脂质含量与成骨细胞凋亡相关性分析。结果白桦脂醇和地塞米松共同作用于小鼠MC3T3-E1细胞后,0.5μmol/L地塞米松(B组)与1.0μmol/L地塞米松组(C组)较空白组(A组),细胞凋亡率、Caspase-3酶活性均明显增高(P<0.05),细胞内甘油三酯含量明显增加。2.0μg/ml白桦脂醇作用细胞后,地塞米松所致的细胞凋亡率上升与Caspase-3酶活性升高受到明显抑制,同时细胞内的甘油三酯蓄积降低。Spearman等级相关分析表明地塞米松诱导后的成骨细胞内脂质含量与细胞凋亡率呈正相关,r=0.412,P<0.05。结论白桦脂醇可显著减轻地塞米松诱导的成骨细胞内脂质蓄积,降低细胞凋亡率;提示地塞米松所致细胞凋亡至少部分与细胞内脂质蓄积有关。
Objective To observe the relationship between endogenous lipid accumulation and apoptosis after the interaction of betulin and dexamethasone in mouse MC3T3-E1 cells. Methods The MC3T3-E1 cells were used as the research object, and the interaction of betulin and dexamethasone was applied to MC3T3-E1 cells to determine the intracellular triglyceride content. The lipid accumulation was observed by NileRed staining. The osteoblast The apoptosis rate of Caspase-3 was assayed by ELISA. Spearman rank correlation was used to analyze the correlation between lipid content and apoptosis of osteoblasts. Results Compared with blank group (group A), 0.5μmol / L dexamethasone (B group) and 1.0μmol / L dexamethasone group (C group), the combination of betulin and dexamethasone combined with MC3T3- The apoptosis rate, Caspase-3 activity were significantly increased (P <0.05), and the intracellular triglyceride content was significantly increased. After treated with 2.0μg / ml betulin, the apoptosis rate induced by dexamethasone and the increase of Caspase-3 activity were significantly inhibited, and the intracellular triglyceride accumulation decreased. Spearman rank correlation analysis showed that the lipid content in osteoblasts induced by dexamethasone was positively correlated with the apoptosis rate (r = 0.412, P <0.05). Conclusion Betulin can significantly reduce the lipid accumulation in osteoblasts induced by dexamethasone and reduce the apoptosis rate. It is suggested that the apoptosis induced by dexamethasone is at least partly related to the accumulation of intracellular lipid.
