脂质过氧化损伤在内毒素休克所致肝细胞损伤的作用越来越引起人们的重视。我室以往研究发现,内毒素休克大鼠肝脏LPO含量升高伴有肝窦内大量白细胞聚集。本文通过观察内毒素、白细胞及两者合用在初代培养肝细胞的脂质过氧化损伤中的作用,试图探讨内毒素休克对肝细胞损伤的机理,并观察中药心脉灵注射液(回阳救急汤的改良剂型)对其的影响。本文按Seglen法,分离大鼠肝细胞,存活率在90%以上,用含胎牛血清DME液调细胞数至1×10~6/ml。将培养的原代肝细胞分成5组,每组5个样本。(1)对照组:正常培养;(2)内毒素组:在肝细胞内加入内
