目的:筛选桂附地黄丸抗中老年男性部分雄激素缺乏综合证(PADAM)的有效部位,并对有效部位的成分来源进行归属分析,为深入研究桂附地黄丸治疗PADAM的药效物质提供基础。方法:(1)制备桂附地黄丸的石油醚、氯仿、乙酸乙酯、正丁醇、水提部位,采用腹腔注射环磷酰胺致大鼠PADAM模型,以大鼠体质量、血清总睾酮(TT)、游离睾酮(FT)、肾脏SOD、MDA为主要评价指标,筛选桂附地黄丸抗PADAM的有效部位;(2)建立有效部位的指纹图谱,并与复方单味药及相应缺味药比较,分析有效部位成分的来源及归属。结果:(1)与正常组比较,模型组大鼠体质量、血清TT、FT含量和肾脏SOD活性显著降低(P<0.01),肾脏MDA水平明显升高(P<0.01)。桂附地黄丸、水提、水+正丁醇、水+乙酸乙酯、水+氯仿、水+石油醚部位能不同程度地改善PADAM大鼠的症状(P<0.01,P<0.05),其中水+正丁醇部位的作用更加显著(P<0.01)。(2)建立了水+正丁醇部位的指纹图谱,确定了以马钱苷为参照物的16个共有峰。(3)初步判断了桂附地黄丸水+正丁醇部位的16个特征峰的归属,此部分化学成分主要来源于熟地黄、牡丹皮、泽泻和山茱萸,以山茱萸所含的特征峰最为丰富。结论:水+正丁醇部位作为桂附地黄丸抗PADAM的有效部位,药效明确,质量可控,实验结果可为进一步研究其药效物质提供实验依据。
OBJECTIVE: To screen the effective parts of anti-androgen-deficient syndrome (PADAM) of Guifu Dihuang Pill in middle-aged and elderly men and to analyze the origin of ingredients of effective parts, so as to provide a basis for further study of Guifu Dihuang Pills in treating PADAM. . Methods: (1) Preparation of Guifu Dihuang Pills petroleum ether, chloroform, ethyl acetate, n-butanol, water extraction site, using intraperitoneal injection of cyclophosphamide rat PADAM model to rat body weight, serum total testosterone TT), free testosterone (FT) and renal SOD and MDA were used as the main evaluation indexes to screen the effective site of anti-PADAM of Guifu Dihuang Pill. (2) The fingerprint of active site was established, Compare and analyze the source and ownership of the effective part of the composition. Results: (1) Compared with normal group, body weight, serum TT, FT contents and renal SOD activity in model group were significantly decreased (P <0.01) and renal MDA levels were significantly increased (P <0.01). Guifu Dihuang Pill, water extract, water + n-butanol, water + ethyl acetate, water + chloroform and water + petroleum ether could ameliorate the symptoms of PADAM rats to some extent (P <0.01, P <0.05) The effect of water + n-butanol fraction was more significant (P <0.01). (2) The fingerprint of water + n-butanol was established, and the 16 common peaks with reference to loganin were identified. (3) The initial identification of the 16 characteristic peaks of water + n-butanol fraction in Guifu Dihuang Pills was mainly attributed to Rehmannia glutinosa, Cortex Moutan, Alisma orientalis and Fructus corni, and the most characteristic peak of Fructus corni rich. Conclusion: The water + n-butanol fraction, as the effective site of anti-PADAM of Guifu Dihuang Pill, has clear pharmacodynamics and controllable quality. The experimental results can provide experimental basis for further study of its pharmacodynamic substances.
