目的:研究大黄影响正常的及内毒素细菌脂多糖(LPS)刺激的大鼠腹腔巨噬细胞(PMφ)分泌肿瘤坏死因子(TNF-α)的特征及其与细胞[Ca~(2+)]_i 的变化关系。方法:利用 TNF-α敏感细胞系 L929,用 MTT法检测 PMφ分泌的 TNF-α量;用荧光探针和荧光光谱法分别测定多细胞和单细胞的[Ca~(2+)]_i。结果:适宜浓度的大黄可轻度活化正常的 PMφ分泌 TNF-α和升高[Ca~(2+)]_i,大黄对 LPS 刺激的 PMφ分泌 TNF-α和升高[Ca~(2+)]_i 有剂量依赖性的抑制效应,1.0%大黄分别抑制了 LPS 刺激的 PMφTNF-α分泌量的92.5%和[Ca~(2+)]_i升高的90.5%。其抑制作用主要表现在给药的早期阶段。结论:大黄对 PMφ分泌 TNF-α有双向调节作用,降低细胞[Ca~(2+)]_i 是其抑制 LPS 刺激作用的细胞内信号传导的关键环节。
Objective: To study the characteristics of tumor necrosis factor-α (TNF-α) secreted by rat peritoneal macrophage (PMφ) stimulated by rhubarb and by lipopolysaccharide (LPS) stimulated by endotoxin and its relationship with cells [Ca 2+ ] The relationship between _i changes. METHODS: The TNF-α sensitive cell line L929 was used to measure the amount of TNF-α secreted by PMφ by MTT assay. The [Ca 2+ ]_i of multicellular and single cells was measured by fluorescence probe and fluorescence spectroscopy, respectively. RESULTS: Rhubarb at a suitable concentration could slightly activate normal PMΦ to secrete TNF-α and increase [Ca 2+ ]_i, Rhubarb to LPS-stimulated PMΦ to secrete TNF-α and increase [Ca 2+ ] __i There was a dose-dependent inhibitory effect. 1.0% Rhubarb inhibited 92.5% of the secretion of PMφTNF-α and 90.5% of [Ca 2+ ]_i, respectively, stimulated by LPS. Its inhibitory effect is mainly manifested in the early stage of drug administration. Conclusion: Rhubarb has two-way regulation of TNF-α secreted by PMφ, and reducing [Ca 2+ ]_i is a key link in intracellular signal transduction that inhibits LPS stimulation.
