目的了解月见草油以不同剂量水平较长期喂养后对受试动物的毒副作用。方法选择清洁级70 g左右的SD大鼠雌雄各40只,随机分为4组,即对照组及三个受试物组,每组雌雄各10只。剂量组分别以1.034、2.069、3.103g/kg.bw剂量的月见草油灌胃90 d,对照组给予等体积植物油。于实验中期和实验末采血测定血液学指标和血生化指标。实验期间观察动物的生长发育情况、不良反应,记录摄食量,计算食物利用率。实验末取肝、肾、脾、睾丸称重后,计算脏器/体重比值;取肝、肾、脾、胃、肠、睾丸、卵巢进行组织病理学检查。结果实验期内大鼠未见明显不良反应。各剂量组大鼠体重增长、进食量、食物利用率、血常规指标、血生化指标、脏器绝对重量、脏器/体重比值与对照组比较,差异均无统计学意义(P>0.05)。大体解剖和组织病理学检查未见与受试样品有关的异常改变。结论在本实验条件下,未见月见草油对受试动物产生明显毒副作用,90 d喂养最大未观察到有害作用剂量为3.103 g/kg.bw。
Objective To understand the toxic side effects of evening primrose oil at different dose levels on the test animals. Methods Forty male and female SD rats with a clean grade of about 70 g were randomly divided into four groups, ie, a control group and three test groups, with 10 females and 10 males in each group. The dose groups were fed with evening primrose oil at doses of 1.034, 2.069, and 3.103 g/kg.bw for 90 days. The control group was given equal volumes of vegetable oil. Blood was measured at the middle and end of the experiment to determine hematological and biochemical parameters. During the experiment, observe the growth and development of animals, adverse reactions, record food intake, and calculate food utilization. At the end of the experiment, the liver, kidney, spleen and testicles were weighed and the organ/body weight ratio was calculated; liver, kidney, spleen, stomach, intestine, testis, and ovary were taken for histopathological examination. Results No significant adverse reactions were observed in rats during the experimental period. The weight gain, food intake, food utilization, blood routine index, blood biochemical index, absolute organ weight, and organ/body weight ratio of each dose group were not statistically different from those of the control group (P>0.05). Gross anatomical and histopathological examinations did not show abnormal changes related to the test sample. Conclusion Under the experimental conditions, there was no obvious toxic side effect of evening primrose oil on the tested animals, and no significant harmful dose of 3.103 g/kg.bw was observed on the 90th day of feeding.
