观察了人重组白细胞介素-13(IL-13)在体外对人红白细胞白血病细胞株(HEL)的作用。该细胞株是巨核细胞白细胞株。培养液中加入或不加入IL-13,HEL细胞培养4天后,计数细胞,然后直接免疫荧光染色,流式细胞仪检测糖蛋白(GP)Ⅱb/Ⅲa表达。为研究IL-13对HEL细胞增殖的影响,细胞培养3天后,用免疫细胞法检测掺入HEL细胞DNA中的5-溴-2′-脱氧尿苷酸(5-Brdu)。结果表明:IL-13使GP Ⅱb/Ⅲa表达增加(与对照组比较,P<0.05),也能促进细胞增殖(与对照组比较,P<0.05)。以上结果提示IL-13对HEL细胞的增殖和分化有显著的调节作用。
The effect of human recombinant interleukin-13 (IL-13) on human erythroblastic leukemia cell line (HEL) was observed in vitro. This cell line is a megakaryocyte white blood cell line. The cells were cultured for 4 days with or without IL-13 in the culture medium. The HEL cells were counted and then directly immunofluorescence staining. The expression of glycoprotein (GP Ⅱb / Ⅲa) was detected by flow cytometry. To investigate the effect of IL-13 on the proliferation of HEL cells, 5-Brdu was incorporated into DNA of HEL cells by immunocytometry after 3 days of cell culture. The results showed that IL-13 increased the expression of GP Ⅱ b / Ⅲ a (P <0.05 compared with the control group), and also promoted cell proliferation (P <0.05 compared with the control group). The above results suggest that IL-13 has a significant regulatory effect on the proliferation and differentiation of HEL cells.
