目的 了解新分离到的禽H9N2亚型流感病毒毒株RNA4节段核苷酸间的关系。方法 病毒RNA经逆转录合成cDNA ,经聚合酶链反应 (PCR)扩增 ,产物纯化 ,采用双脱氧链末端终止法进行核苷酸序列测定。结果 香港鹌鹑流感病毒 (quHK197)、香港猪流感病毒 (swHK998)、香港鸡流感病毒 (ckHK997)和禽代表株威斯康星火鸡流感病毒(tywis6 6 )的H9N2亚型RNA4节段长度分别为 16 72、16 70、16 5 1和 16 83个核苷酸 ,编码血凝素 (HA)蛋白。这 3株分离到的禽H9N2亚型之间的同源性在 92 7%~ 96 9%之间 ,与禽H9N2代表株间的同源性在 87 8%~ 89 0 %之间。结论 新分离到的禽H9N2亚型毒株与代表株的同源性差异较大。
Objective To understand the relationship between nucleotide sequences of RNA4 segments of newly isolated avian influenza virus H9N2 strain. Methods The viral RNA was reverse transcribed into cDNA and amplified by polymerase chain reaction (PCR). The product was purified and the nucleotide sequence was determined by dideoxy chain termination method. Results The lengths of H4N2 subtype RNA4 segment of quail influenza virus (quHK197), swine influenza (swHK998), chick chicken influenza virus (ckHK997) and avian influenza virus (tywis6 6) of Hong Kong were 16 72, 16 70, 16 5 1 and 16 83 nucleotides encoding the hemagglutinin (HA) protein. The identities of the H9N2 isolates isolated from these three isolates ranged from 92.7% to 96.9%, and their homologies with the avian H9N2 strain ranged from 87.8% to 89.0%. Conclusion The homology of newly isolated H9N2 avian strains and representative strains is quite different.
