目的 探讨γ干扰素受体α(interforon gamma receptor alpha IFN-γRα)及细胞间黏附分子I(intercelluar adhesion moleeule I.ICAM-1)在大鼠肝卵圆细胞增殖模型中的表达情况及其生物学意义。 方法 采用二乙酰氨基笏(2-acetaminofluorene AAF)加三分之二肝切建立大鼠肝卵圆细胞增殖的模型,并以大鼠单纯三分之二肝切后的再生肝纠织、单纯灌AAF4天后的肝组织及正常大鼠肝细胞作对照,采用RT-PCR检测IPN-γRα、ICAM-1及甲胎蛋白(alpha-fetoprotein AFP)mRNA在模型组及对照组肝组织中的表达情况。结果 AFP,IFN-γRα及ICAM-1mRNA 表达水平在模型组肝切后4天及9天明显上调,AFP在肝切后9天、IFN-γRa及ICAM-1在肝切后4天的表达均显著高于对照组(P<0.05),在肝切后20天复常。结论 AFN-γRα和ICAM-1mRNA的高表达与肝卵圆细胞的增殖及分化过程密切相关。
Objective To investigate the expression of interforon gamma receptor alpha (IFN-γRα) and intercellular adhesion molecule I (ICAM-1) in rat hepatic oval cell proliferation model and its biology significance. Methods A rat model of hepatic oval cell proliferation was established by 2-acetaminofluorene (AAF) plus two-thirds hepatectomy. The rats were sacrificed by simple two-thirds hepatectomy, AAF4 days after liver tissue and normal rat liver cells as a control, RT-PCR detection of IPN-γRα, ICAM-1 and alpha-fetoprotein (AFP) mRNA expression in the model group and control group liver tissue. Results The levels of AFP, IFN-γRα and ICAM-1 mRNA were significantly increased 4 and 9 days after hepatectomy in model group. The expression of AFP, IFN-γRa and ICAM-1 at 4 days after hepatectomy Significantly higher than the control group (P <0.05), normalization 20 days after hepatectomy. Conclusion The high expression of AFN-γRα and ICAM-1 mRNA is closely related to the proliferation and differentiation of hepatic oval cells.
