目的观察不同剂量姜黄素对HaCaT细胞中FGF10 mRNA转录及其蛋白翻译的影响。方法HaCaT细胞经不同剂量的姜黄素处理48h后,采用免疫荧光和原位杂交的方法检测FGF10 mRNA转录及其蛋白翻译的改变。结果与未处理组比较,处理48h后,FGF10 mRNA转录及其蛋白翻译水平随姜黄素剂量的增加而减少,两组间比较有显著性差异(P<0.05)。结论姜黄素可抑制HaCaT细胞中FGF10 mRNA转录及其蛋白翻译,并随姜黄素剂量的增加,抑制作用加强。
Objective To observe the effects of different doses of curcumin on transcription and protein translation of FGF10 mRNA in HaCaT cells. Methods HaCaT cells were treated with different doses of curcumin for 48 hours. The changes of FGF10 mRNA transcription and protein translation were detected by immunofluorescence and in situ hybridization. Results Compared with the untreated group, after 48 hours of treatment, the transcription of FGF10 mRNA and its protein translation decreased with the increase of curcumin dose. There was a significant difference between the two groups (P<0.05). Conclusion Curcumin can inhibit the transcription and protein translation of FGF10 mRNA in HaCaT cells, and the inhibitory effect is enhanced with the increase of curcumin dose.
