采用白细胞介素-6(Interleukin6,IL-6)依赖细胞株7TD1及MTT比色法,动态观察了正常及内毒素注射后不同时间大鼠血清和支气管肺泡灌洗液(bronchoalveolarlavagefluid,BALF)中IL-6含量的变化;同时,还对各组大鼠的肺体指数和BALF中蛋白质含量进行了测定。结果显示,大鼠注射内毒素后1、3、6、12h,其血清和BALF中IL-6含量急剧升高(P<0.01),注射后24h降为正常(P>0.05);此外,其肺体指数和BALF中蛋白质含量亦明显高于对照组,尤以3h和6h显著(P<0.01);且内毒素致伤后大鼠血清和BALF中IL-6含量的升高与BALF中蛋白质含量的升高呈显著正相关。推测IL-6可能参与了肺水肿的形成,其机制有待进一步探讨.
Interleukin 6 (IL-6) -dependent cell line 7TD1 and MTT assay were used to observe the changes of IL-6 in serum and bronchoalveolar fluid (BALF) at different time points after normal and endotoxin injection -6 content; at the same time, the lung index and the protein content of BALF in each group were also measured. The results showed that the levels of IL-6 in serum and BALF increased sharply at 1, 3, 6 and 12 h after injection of endotoxin (P <0.01), and decreased to normal at 24 h after injection (P> 0.05) In addition, the lung index and BALF protein content were also significantly higher than the control group, especially at 3h and 6h (P <0.01); and endotoxin-induced serum and BALF IL-6 content There was a significant positive correlation between the increase and protein content in BALF. Speculated that IL-6 may be involved in the formation of pulmonary edema, the mechanism needs further study.
