以相应引物对日本囊对虾(Marsupenaeus japonicus)线粒体DNA细胞色素氧化酶I亚基基因(mtDNA CO I)进行PCR扩增,经过基因重组、转化、克隆、筛选、DNA测序,得到709 bp的碱基片断.碱基组成A、C、G、T含量分别为196bp(27.64%)、129 bp(18.19%)1、34 bp(18.90%)、250 bp(35.26%).与GenBank中斑节对虾(Penaeus monodon)的mtD-NA CO I全序列(AF217843)比对,经分析发现:本实验获得的日本囊对虾mtCO I基因片段序列和GenBank上的同种序列(AY264897)都只是该种CO I基因序列的一部分,二者之间有41 bp的重叠并可拼接,拼接后的总长为1 515 bp.
The mtDNA COI of mitochondrial DNA of Marsupenaeus japonicus was amplified by PCR from the corresponding primers. After the recombination, transformation, cloning, screening and DNA sequencing, a 709 bp base (A, C, G and T) were 196bp (27.64%), 129 bp (18.19%), 1,34 bp (18.90%) and 250 bp (35.26% (AF217843) of Penaeus monodon, we found that the mtCO I gene fragment and the homologous sequence in GenBank (AY264897) obtained from this experiment are only the COI gene A portion of the sequence, with a 41 bp overlap between the two, can be spliced, with a total length of 1 515 bp.
