目的 探讨 β -七叶皂甙钠对单侧睾丸扭转 /复位后双侧睾丸细胞凋亡及相关基因蛋白表达的影响。 方法 3 0只青春期Wistar大鼠随机等分为对照组 (C组 )、扭转 /复位模型组 (M组 ) (左侧睾丸扭转 72 0°、2h后复位 )和 β -七叶皂甙钠治疗组 (T组 )。术后 48h获取双侧睾丸 ,分别以原位缺口末端标记法 (TUNEL)及免疫组织化学法检测凋亡细胞和bcl -2、Bax基因蛋白表达。结果 与C组比较 ,M组双侧睾丸生精细胞凋亡指数明显增加、bcl -2表达下降和Bax表达增强 (P <0 0 1) ,bcl -2 /Bax比值降低。与M组比较 ,T组双侧睾丸生精细胞凋亡指数明显减少 (P <0 0 1)、病理损害减轻 ,Bax低表达 (P <0 0 1) ,bcl -2 /Bax比值增高。结论 单侧睾丸扭转复位后 ,双侧睾丸生精细胞凋亡加剧 ,bax、bcl -2参与了细胞凋亡的调控 ;β -七叶皂甙钠通过抑制bax的表达 ,提高bcl -2 /Bax比值 ,逆转生精细胞凋亡增加而发挥保护作用。
Objective To investigate the effect of β - aescinate on the apoptosis of testis cells and the expression of related genes after unilateral testicular torsion / detorsion. Methods Twenty - three adolescent Wistar rats were randomly divided into control group (C group), untreated group (M group) (left testicular torsion 72 0 °, reduction 2 h) and β - aescinate treatment group (T group). Bilateral testes were harvested 48h after operation. Apoptotic cells and expressions of bcl-2 and Bax protein were detected by TUNEL and immunohistochemistry respectively. Results Compared with group C, the apoptosis index of spermatogenic cells in testis of M group was significantly increased, the expression of bcl - 2 and Bax increased (P <0.01), and the ratio of bcl -2 / Bax decreased. Compared with M group, the apoptosis index of spermatogenic cells in testis of group T was significantly decreased (P <0.01), the pathological damage was alleviated, the expression of Bax was low (P <0.01), and the ratio of bcl -2 / Bax was increased. Conclusion Apoptosis of spermatogenic cells in bilateral testes is intensified after unilateral testicular torsion and reduction, and bax and bcl -2 are involved in the regulation of apoptosis. Β - aescinate enhances the ratio of bcl -2 / Bax by inhibiting the expression of bax , Reversing the apoptosis of spermatogenic cells to play a protective effect.
