目的 测定大鼠内毒素血症期间淋巴细胞释放和合成降钙素基因相关肽 (CGRP)的变化。方法 采用逆转录聚合酶链反应 (RT- PCR)测定大鼠淋巴细胞 CGRP m RNA水平。用放射性标记法测定血浆 CGRP水平。结果 注射内毒素大鼠血浆和淋巴细胞释放 CGRP均增高 ,淋巴细胞中 CGRP m RNA表达也明显增高。结论 提示内毒素不仅刺激淋巴细胞释放 CGRP,而且能通过某些机制激活淋巴细胞 CGRP m RNA的转录 ,使 CGRP合成增加 ,以作为 CGRP大量释放的重要补充来源。
Objective To determine the changes of lymphocyte release and synthesis of calcitonin gene related peptide (CGRP) during endotoxemia in rats. Methods The levels of CGRP-m RNA in rat lymphocytes were determined by reverse transcription-polymerase chain reaction (RT-PCR). Plasma CGRP level was measured by radiolabeling. Results The levels of CGRP released from plasma and lymphocytes of endotoxin-treated rats were significantly increased, and the expression of CGRP-m RNA in lymphocytes was also significantly increased. The results suggest that endotoxin not only stimulates the release of CGRP from lymphocytes, but also activates the transcription of CGRP-m RNA in lymphocytes by some mechanisms and increases the synthesis of CGRP as an important supplementary source for the release of large amounts of CGRP.
