以中国李Nubiana试管苗幼嫩叶片上的叶柄为试材,研究了基本培养基、激素种类配比、暗培养时间、培养基中琼脂浓度等对叶柄再生的影响,建立了其叶柄再生体系。结果表明,中国李Nubiana叶柄再生不定芽最佳培养基为WPM+TDZ 0.5 mg.L-1+蔗糖30 g.L-1+琼脂6 g.L-1,不定芽再生率可达到35.0%;暗培养为叶柄不定芽再生的必要条件,暗培养14 d后转入光下培养,能促进叶柄产生不定芽。中国李Nubiana叶柄再生表现出明显的极性,不定芽一般出现在叶柄的近轴端。
The petiole on young leaves of young Chinese Nubiana in vitro was used as experimental material to study the effect of basic medium, hormone type, dark culture time and agar concentration in medium on petiole regeneration, and the petiole regeneration system was established. The results showed that the optimum culture medium for adventitious buds regeneration from petiole of Chinese Nubiana was WPM + TDZ 0.5 mg.L-1 + sucrose 30 gL-1 + agar 6 gL-1, and the adventitious bud regeneration rate could reach 35.0% Adventitious bud regeneration of the necessary conditions, dark culture after 14 days into the light culture, can promote adventitious buds produce petiole. Petiole regeneration of Chinese plum Nubiana showed obvious polarity, and adventitious buds generally appeared on the paraxial side of petioles.
