目的:对中国南海桶形芋螺(Conus betulinus)毒液的化学成分进行研究。方法:采用凝胶色谱、高效液相色谱对桶形芋螺毒液分离纯化,得到一个肽类毒素,通过质谱分析、氨基酸组成分析、N-端序列分析、二硫键定位确定了其化学结构,用合成物对化学结构进行了验证。结果:从中国南海桶形芋螺毒液中鉴定了1个芋螺毒素,氨基酸序列为H-Ser-Gla-Cys-Cys-Ile-Arg-Asn-Phe-Leu-Cys-Cys-OH,其中Cys3与Cys10,Cys4与Cys11形成2个链内二硫键,命名为芋螺毒素bt5a。结论:芋螺毒素bt5a为未见文献报道的新毒素,是芋螺毒素T-超家族的一个新成员。
Objective: To study the chemical constituents of Conus betulinus venom in the South China Sea. METHODS: The peptide was purified by gel chromatography and high performance liquid chromatography (HPLC) to obtain a peptide toxoid. Its chemical structure was confirmed by mass spectrometry, amino acid composition analysis, N-terminal sequence analysis and disulfide bond localization. The chemical structure was verified with the composition. Results: One conotoxin was identified from the conchocerone in the South China Sea. The amino acid sequence was H-Ser-Gla-Cys-Cys-Ile-Arg-Asn-Phe-Leu-Cys-Cys- With Cys10, Cys4 and Cys11 form two intrachain disulfide bonds, named conotoxin bt5a. CONCLUSION: Conotoxin bt5a is a new toxin not reported in the literature and is a new member of the conotoxin T-superfamily.
