目的:探讨细胞内Fas相关死亡域样白介素1-β转换酶抑制蛋白(FLIP)在胃癌组织中的表达及其对胃癌细胞凋亡的影响。方法:荧光定量逆转录聚合酶链反应(FQ-RT-PCR)检测胃癌组织及其癌旁正常组织FLIP(S)mRNA表达水平。结果:FLIP(S)mRNA在胃癌组织和癌旁正常组织都有表达,在胃癌组织表达水平明显高于癌旁正常组织(P<0.05)。FLIP(S)mRNA在低分化癌组织中表达较中高分化表达高,有淋巴结转移表达高于无淋巴结转移,随着浸润深度的增加,FLIP(S)mRNA表达也增加,差异有统计学意义(P<0.05)。cFLIP的蛋白表达与胃癌细胞凋亡指数呈负相关(P<0.05)。结论:FLIP(S)在胃癌组织中表达升高,在分化越低组织中其表达水平越高,抑制胃癌癌细胞的凋亡,促进胃癌的发生,为临床治疗胃癌提供了新思路。
Objective: To investigate the expression of intracellular Fas-related death domain-like interleukin 1-β converting enzyme inhibitory protein (FLIP) in gastric cancer and its effect on gastric cancer cell apoptosis. Methods: The expression of FLIP (S) mRNA in gastric cancer tissue and adjacent normal tissues was detected by fluorescence quantitative reverse transcription polymerase chain reaction (FQ-RT-PCR). Results: The expression of FLIP (S) mRNA in gastric cancer tissues and adjacent normal tissues was significantly higher than that in adjacent normal tissues (P <0.05). The expression of FLIP (S) mRNA in poorly differentiated carcinoma tissues was higher than that in moderately well differentiated ones, and higher in lymph node metastasis than in those without lymph node metastasis. The expression of FLIP (S) mRNA also increased with the depth of invasion P <0.05). The protein expression of cFLIP was negatively correlated with the apoptosis index of gastric cancer cells (P <0.05). Conclusion: The expression of FLIP (S) in gastric cancer tissue is higher than that in the lower differentiated tissues, which inhibits the apoptosis of gastric cancer cells and promotes the occurrence of gastric cancer. It provides a new idea for the clinical treatment of gastric cancer.
