目的分析滨州地区供血者ABO亚型的分子机制、分布情况。方法采用试管法对微板法筛查出的正反定型不符血样做进一步血型血清学分析,并进行亚型确认。对亚型标本进行血清学分析和DNA测序,测序结果用软件比对分析后,确定其基因型。结果从2014~2015年滨州地区无偿献血者84 394份血样中检出ABO亚型12例(检出率为1.4/万),其中A亚型3例(A~(el)1例、A_31例、A~(el)B1例)占25%,B亚型8例(B~(el)1例、Bx1例、B_32例、AB_34例)占66.7%,B(A)表型1例占8.3%。对12例亚型标本进行DNA测序结果显示,Ael05基因2例、B301基因1例、B303基因4例、B(A)04基因1例,但有A亚型1例、B亚型3例,6、7外显子测序未见明显变化。结论 ABO亚型分布存在不均衡性和地域性差异,滨州地区最常见的ABO亚型为AB3或B3,最常见突变基因为突变发生在第3内含子的B303基因。
Objective To analyze the molecular mechanism and distribution of ABO subtype in blood donors in Binzhou area. Methods The test-tube method was used for further blood group serological analysis on positive and negative unscientific blood samples screened by microplate method and confirmed by subtype. Serotyping and DNA sequencing of subtype specimens were performed. The sequencing results were analyzed by software and the genotypes were determined. Results 12 cases of ABO subtypes were detected in 84 394 blood samples of unpaid blood donors in Binzhou area from 2014 to 2015 (the detection rate was 1.4 / million). Among them, 3 cases of A subtypes (A ~ (el), A ~ 31 cases , B1 (25 cases), 8 cases of B subtype (1 case of B el, Bx1 case, B_32 case, AB_34 case) accounted for 66.7%, while B (A) %. DNA sequencing of 12 subtypes showed that there were 2 cases of Ael05 gene, 1 case of B301 gene, 4 cases of B303 gene and 1 case of B (A) 04 gene, but there were 1 case of type A subtype and 3 cases of type B subtype. 6,7 exon sequencing no significant change. Conclusions The distribution of ABO subtypes is uneven and regional. The most common ABO subtype in Binzhou is AB3 or B3, and the most common mutation is B303 with mutation in the third intron.
