为观察芩丹颗粒对热损伤大鼠心功能的保护作用,将75只健康清洁级Wistar雄性大鼠随机分为5组,分别为常温对照(生理盐水)组、热损伤模型组和10、20、40 g/kg芩丹颗粒组,每组15只。热暴露前,采用灌胃方式进行给药(热损伤模型组给予生理盐水),每天1次,连续给药20 d;然后将模型组和各剂量芩丹颗粒组大鼠暴露于43℃热舱中一次性暴露70 min。实时监测体核温度(Tc)、平均动脉压(MAP)、心率(HR)的变化,检测大鼠血清白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)含量和乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)活力,并观察心肌组织的损伤程度。结果显示,与常温对照组比较,热损伤模型组大鼠的Tc升高,而MAP和心率降低;血清中IL-1和TNF-α的含量和LDH、CK、AST、ALP的活力及心肌组织坏死和血管充血的评分均升高,差异均有统计学意义(P<0.05)。与热损伤模型组比较,仅40 g/kg芩丹颗粒组大鼠的Tc降低,而MAP和心率升高;血清中IL-1和TNF-α的含量均降低及心肌组织坏死和血管充血的评分均降低;20、40 g/kg芩丹颗粒组大鼠血清中LDH、CK、AST、ALP的活力均较高,除20g/kg芩丹颗粒组AST活力外,差异均有统计学意义(P<0.05)。且随着芩丹颗粒给药剂量的升高,大鼠的Tc呈下降趋势,而MAP和心率均呈上升趋势;血清中IL-1和TNF-α的含量及LDH、CK、AST、ALP的活力以及心肌组织坏死和血管充血的评分均呈下降趋势。提示芩丹颗粒可能通过减轻炎症反应对热应激大鼠心功能起到保护作用。
