采用修改Hamburger-Salmon体外双层琼脂法与琼脂培养MTT法同时对人肺癌细胞进行药物敏感性测定。结果表明:琼脂培养MTT法成功率为100%(42/42);双层琼脂法成功率为50%(21/42)。两法同时可供药敏分析的126药物浓度次中,琼脂培养MTT法测定肺癌细胞的药物敏感率为32.5%(41/126),双层琼脂法的药物敏感率为39.7%(50/126),两者间无明显差异(p>0.05);两法测定结果的总符合率为92.8%(117/126)。提示琼脂培养的MTT法测定可以取代双层琼脂法测定人实体瘤对不同药物的反应以供临床参考。
Modified Hamburger-Salmon in vitro double-layer agar assay and agar culture MTT assay were used to simultaneously determine the drug susceptibility of human lung cancer cells. The results showed that the success rate of MTT assay in agar culture was 100% (42/42); the success rate of double-layer agar assay was 50% (21/42). The drug sensitivity rate of lung cancer cells was 32.5% (41/126) in the agar culture MTT assay, and 39.7% in the double-layer agar assay. (50/126), there was no significant difference between the two (p>0.05); the overall compliance rate of the two methods was 92.8% (117/126). It is suggested that the MTT assay for agar culture may replace the double-layer agar assay for the determination of human solid tumor response to different drugs for clinical reference.
