应用Ｎｏｒｔｈｅｒｎ印迹方法研究高血压（ＳＨＲ）和正常（ＷＫＹ）大鼠脑微血管培养内皮细胞，在剪切应力０、０．５、１、２Ｐａ（１Ｐａ＝１０ｄｙｎ／ｃｍ２）作用２４ｈ后检测内皮素及其基因ｍＲＮＡ表达上的区别。结果表明：在剪切应力０、０．５、１Ｐａ下，ＷＫＹ大鼠的内皮细胞随着剪切应力加大，其内皮素及其基因ｍＲＮＡ表达水平提高；ＳＨＲ大鼠的内皮细胞也随着剪切应力加大而加大，但其内皮素水平及其基因的ｍＲＮＡ表达均比ＷＫＹ相应组的为高。在剪切应力２Ｐａ时，ＷＫＹ和ＳＨＲ大鼠的内皮细胞内皮素及其基因的ｍＲＮＡ表达水平不再继续提高；但ＳＨＲ组内皮素水平及其基因的ｍＲＮＡ表达仍比ＷＫＹ相应组的为高。表明ＷＫＹ和ＳＨＲ两组大鼠内皮素及其基因的ｍＲＮＡ表达对于剪切应力在某种范围内具有正相关性，而超出某种范围则没有这种相关性，其中ＳＨＲ组大鼠的内皮素水平及其基因ｍＲＮＡ表达不论剪切应力多大，均处于较ＷＫＹ组高的表达状态。为ＳＨＲ大鼠对于剪切应力引起的血管收缩功能的增强、舒张功能的减弱、血管重建能力下降和易产生动脉硬化损伤提供新思路。

The results showed that the mRNA expression of endothelin and its gene in WKY and SHR groups had a positive correlation with the shear stress in a certain range, while beyond that range, there was no such correlation. Among them, the endothelin Regardless of the magnitude of the shear stress, the level of mRNA expression was higher than that of the WKY group. It provides a new idea for SHR rats to enhance the function of vasoconstriction induced by shear stress, reduce the diastolic function, reduce the ability of vascular remodeling and easy to produce atherosclerosis damage.
