应用Northern印迹方法研究高血压(SHR)和正常(WKY)大鼠脑微血管培养内皮细胞,在剪切应力0、0.5、1、2Pa(1Pa=10dyn/cm2)作用24h后检测内皮素及其基因mRNA表达上的区别。结果表明:在剪切应力0、0.5、1Pa下,WKY大鼠的内皮细胞随着剪切应力加大,其内皮素及其基因mRNA表达水平提高;SHR大鼠的内皮细胞也随着剪切应力加大而加大,但其内皮素水平及其基因的mRNA表达均比WKY相应组的为高。在剪切应力2Pa时,WKY和SHR大鼠的内皮细胞内皮素及其基因的mRNA表达水平不再继续提高;但SHR组内皮素水平及其基因的mRNA表达仍比WKY相应组的为高。表明WKY和SHR两组大鼠内皮素及其基因的mRNA表达对于剪切应力在某种范围内具有正相关性,而超出某种范围则没有这种相关性,其中SHR组大鼠的内皮素水平及其基因mRNA表达不论剪切应力多大,均处于较WKY组高的表达状态。为SHR大鼠对于剪切应力引起的血管收缩功能的增强、舒张功能的减弱、血管重建能力下降和易产生动脉硬化损伤提供新思路。

The results showed that the mRNA expression of endothelin and its gene in WKY and SHR groups had a positive correlation with the shear stress in a certain range, while beyond that range, there was no such correlation. It provides a new idea for SHR rats to enhance the function of vasoconstriction induced by shear stress, reduce the diastolic function, reduce the ability of vascular remodeling and easy to produce atherosclerosis damage.
