目的探讨层粘连蛋白(Laminin,LN)在大鼠脑缺血再灌注中的作用。方法制备大鼠脑缺血再灌注(MCAO/R)模型,分为假手术组与缺血再灌注组;缺血2h后依照再灌注时间点的不同分为2h、6h、12h、24h、3d及7d共6组。各时相点取材,免疫组化观察脑组织LN的表达变化,并使用多媒体彩色病理图像分析系统进行定量分析。结果LN的表达于再灌注2h开始降低,24h时达最低,第3日时开始回升。结论LN可能参与了缺血再灌注血管源性脑水肿的形成与发展,并且可能参与了再灌注损伤后新生血管的生成、血管重塑及侧支循环的建立。
Objective To investigate the role of laminin (LN) in cerebral ischemia-reperfusion in rats. Methods The rat model of cerebral ischemia-reperfusion (MCAO / R) was established and divided into sham-operated group and ischemia-reperfusion group. The rats were divided into 2h, 6h, 12h, 24h, 3d And 7d a total of 6 groups. At each time point, the expression of LN in the brain tissue was observed by immunohistochemistry and quantitatively analyzed by using the multimedia color pathological image analysis system. Results The expression of LN began to decrease at 2h after reperfusion, reached the lowest at 24h and started to rebound on the third day. Conclusion LN may be involved in the formation and development of vasogenic brain edema induced by ischemia and reperfusion, and may be involved in the formation of neovascularization, vascular remodeling and collateral circulation after reperfusion injury.
