Kozbo 等人首次建立人T-B 细胞杂交瘤。但其融合细胞均系肿瘤细胞,T 细胞系SUP-T_1来源于淋巴白血病细胞,B 细胞系GM-1500来源于骨髓细胞。作者用正常T 细胞与正常B 淋巴母细胞样细胞系729-HF 成功构建了人T-B 细胞杂交瘤。T 细胞融合前先体外激活;选择的729-HF B 细胞是EBV 转染的细胞.产生Ig 能力很弱,敏感的ELISA 仅能测到表面少量IgM。融合后筛选出三株细胞,研究表面标志、有关基因和功能。杂交瘤细胞同时表达了T 细胞表而标志CD3、CD8、CD2和B 细胞
Kozbo et al. first established human T-B cell hybridomas. However, the fusion cells were all tumor cells. The T cell line SUP-T_1 was derived from lymphoid leukemia cells and the B cell line GM-1500 was derived from bone marrow cells. The authors successfully constructed human T-B cell hybridomas with normal T cells and normal B lymphoblastic cell line 729-HF. The T cells were activated in vitro prior to fusion; the selected 729-HF B cells were EBV-transfected cells. The ability to produce Ig was weak, and sensitive ELISA could detect only small amounts of surface IgM. Three cells were screened after fusion to study surface markers, related genes and functions. Hybridoma cells simultaneously express the T cell table and mark CD3, CD8, CD2, and B cells
