目的 观察急性一氧化碳中毒 (ACMP)后小鼠血小板膜糖蛋白 CD31、CD6 1和 CD6 2 p的变化及高压氧 (HBO)对其影响。方法 雄性昆明小鼠共 4 2只随机分成 7组 ,正常对照组、HBO(ACMP后 HBO治疗 )第 1、3、5天组及 ACMP第 1、3、5天组 ,每组各 6只。用流式细胞仪测定血小板膜上 CD31、CD6 1的平均荧光强度及 CD6 2 p的阳性率。结果 ACMP第 1天组和 HBO第 1天组 CD31的表达水平均明显高于正常对照组 (P<0 .0 1)。 ACMP第 1、3、5天组和 HBO第 1、3、5天组 CD6 1的表达水平均明显高于正常对照组 (P<0 .0 5或 P<0 .0 1) ;HBO第 3天组 CD6 1的表达水平明显低于同天 ACMP组 (P<0 .0 5 )。 ACMP第 1、3天组和 HBO第 3天组 CD6 2 p的表达水平均明显高于正常对照组 (P<0 .0 5 ) ;HBO第 1、3天组 CD6 2 p的表达水平均明显低于同天 ACMP组 (P<0 .0 5 )。结论 ACMP可显著增加CD31、CD6 1和 CD6 2 p在血小板膜上的表达 ,提示血小板活化反应增强 ;早期行 HBO治疗可显著减少CD6 1和 CD6 2 p的表达水平 ,而 CD31的表达水平无显著改变。
Objective To observe the changes of platelet membrane glycoproteins CD31, CD6 1 and CD6 2 p in mice after acute carbon monoxide poisoning (ACMP) and the effects of hyperbaric oxygen (HBO) on them. Methods A total of 42 male Kunming mice were randomly divided into 7 groups: normal control group, HBO group (1st, 3rd, 5th, 5th day after ACMP) and ACMP groups 1, 3 and 5, 6 in each group. The average fluorescence intensity of CD31 and CD6 1 on platelet membrane and the positive rate of CD6 2 p were measured by flow cytometry. Results The expression of CD31 on day 1 of ACMP and day 1 of HBO were significantly higher than that of the normal control group (P <0.01). The expression of CD6 1 in the 1st, 3rd, 5th day of ACMP and the 1st, 3rd and 5th day of HBO group were significantly higher than that of the normal control group (P <0.05 or P <0.01) The expression level of CD6 1 in day group was significantly lower than that in ACMP group (P <0.05). The expression of CD6 2 p in ACMP day 1, day 3 group and HBO day 3 group were significantly higher than those in normal control group (P <0.05) Lower than the same day ACMP group (P <0. 05). Conclusion ACMP can significantly increase the expression of CD31, CD6 1 and CD6 2 p on the platelet membrane, suggesting that the platelet activation reaction is enhanced. The early HBO treatment can significantly reduce the expression of CD6 1 and CD6 2 p, while the expression level of CD31 is not significant change.
