目的:研究三七素对谷氨酸损伤的PC12细胞的影响。方法:用谷氨酸复制体外培养的PC12细胞损伤模型,采用MTT法、Hoechst33342/PI双重染色法分别研究高、低剂量三七素对谷氨酸所致的PC12细胞损伤的影响。结果:谷氨酸刺激后,PC12细胞存活率较对照组显著降低(P<0.05)、凋亡显著增强(P<0.05)。给予高剂量三七素可加重谷氨酸引起的PC12增殖力降低、凋亡增强。但低剂量三七素干预后,细胞存活率较模型组显著升高(P<0.05),凋亡较模型组明显降低(P<0.05)。结论:高剂量三七素可加重谷氨酸对PC12细胞的损伤,但低剂量三七素可显著减轻谷氨酸对PC12细胞的损伤,但具体机制尚有待于进一步研究。
Objective: To study the effect of Panax notoginseng on glutamate - induced PC12 cells. Methods: The injury model of PC12 cells cultured in vitro was induced by glutamate. MTT assay and Hoechst33342 / PI double staining were used to study the effects of high and low doses of PNS on the injury of PC12 cells induced by glutamate. Results: After glutamate stimulation, the survival rate of PC12 cells was significantly lower than that of the control group (P <0.05), and the apoptosis was significantly increased (P <0.05). Given a high dose of three seven elements can increase glutamate-induced PC12 decreased proliferation, increased apoptosis. However, the cell survival rate was significantly increased (P <0.05) and the apoptosis rate was significantly lower than that of the model group (P <0.05). CONCLUSION: High dose of Panax notoginseng can aggravate the damage of PC12 cells induced by glutamate, but the low dose of Panax notoginseng can significantly reduce the injury of PC12 cells by glutamate, but the specific mechanism remains to be further studied.
