目的:研究杨桃根提取物(YTG)对链脲佐菌素(STZ)致糖尿病小鼠的降糖作用。方法:SPF级KM雄性小鼠60只,采用尾静脉注射STZ建立糖尿病小鼠模型,将成模小鼠随机分成模型组,二甲双胍组(0.25 g·kg-1·d-1),YTG高、中、低剂量组(1.2,0.6,0.3 g·kg-1·d-1),另设正常组,每组10只,ig给药4周,在第2,4周末测定空腹血糖(FBG),实验末期进行糖耐量试验(OGTT),并测定肝组织中肝糖原含量,己糖激酶(HK),丙酮酸激酶(PK),超氧化物歧化酶(SOD),丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)的水平。结果:与正常组比较,模型组给药FBG明显升高,血糖曲线下面积明显升高,肝糖原,HK,PK水平明显降低,肝组织中MDA含量明显升高,SOD,GSH-Px水平明显降低,均具有明显统计学差异(P<0.01);与模型组比较,各YTG给药组,FBG明显下降,血糖曲线下面积明显降低,肝糖原含量增加,HK,PK活性增加,肝组织中SOD和GSHPx活性明显升高,MDA含量降低,均具有统计学差异(P<0.05,P<0.01)。结论:杨桃根提取物能降低糖尿病小鼠的血糖,提高糖耐量,其机制可能与增强HK,PK的活性,提高肝脏的抗氧化能力有关。
Objective: To study the hypoglycemic effect of Caragana root extract (YTG) on streptozotocin (STZ) -induced diabetic mice. Methods: Sixty SPF KM male mice were randomly divided into model group, metformin group (0.25 g · kg -1 · d -1), high YTG , Low dose group (1.2,0.6,0.3 g · kg-1 · d-1), another normal group, 10 rats in each group, which were administered for 4 weeks. Fasting blood glucose (FBG) Glucose tolerance test (OGTT) was performed at the end of the experiment. Hepatic glycogen content, hexokinase (HK), pyruvate kinase (PK), superoxide dismutase (SOD), malondialdehyde (MDA) Glutathione peroxidase (GSH-Px) levels. Results: Compared with the normal group, FBG in the model group was significantly increased, the area under the blood glucose curve was significantly increased, liver glycogen, HK, PK levels were significantly decreased, the content of MDA in liver tissue was significantly increased, the levels of SOD, GSH-Px (P <0.01). Compared with the model group, the levels of FBG in each YTG-treated group decreased significantly, the area under the blood glucose curve decreased significantly, the content of hepatic glycogen increased, the activity of HK and PK increased, and the liver The activities of SOD and GSHPx in the tissues were significantly increased and the content of MDA was decreased, with statistical significance (P <0.05, P <0.01). Conclusion: Carambola extract can reduce the blood sugar of diabetic mice and improve the glucose tolerance. The mechanism may be related to the enhancement of the activity of HK and PK, and the enhancement of the anti-oxidative capacity of the liver.
