采用离体肠管灌流法,探讨了神经降压素(NT)对家兔离体空、回肠平滑肌收缩的影响及其可能机制.结果表明:①微量NT即可抑制家兔离体空、回肠平滑肌的收缩;在空肠,短暂的抑制期过后代之以收缩幅度增大,但基线不变;在回肠则为持续约1min的收缩抑制反应.NT对空、回肠平滑肌收缩的影响皆具有明显的量效依赖关系.②纳洛酮几乎完全消除NT引起的空肠平滑肌的抑制后收缩增强效应(P<0.01),但不影响NT对空、回肠平滑肌的收缩抑制效应(P>0.05).③河豚毒素(TTX)、酚妥拉明和心得安皆不影响NT对空、回肠平滑肌收缩的效应(P>0.05).结果提示NT可能直接作用于空、回肠平滑肌膜上的NT敏感受体;空肠平滑肌的抑制后收缩增强效应与阿片肽及其受体相关.
The effect of neurotensin (NT) on contractile activity of isolated empty and ileum smooth muscle of rabbits and its possible mechanism were explored by ex vivo intestinal perfusion method.The results showed that: (1) Micro-NT could inhibit the in vitro empty and ileum smooth muscle In the jejunum, shortening of the period after the suppression of the generation to increase the amplitude of the contraction, but the same baseline; in the ileum for about 1min contraction inhibition response.NT on the empty and ileum smooth muscle contraction have a significant amount of (P <0.01), but did not affect the contractile inhibitory effect of NT on the contractility of empty and ileum smooth muscles (P> 0.05) .③Toxicity of tetrodotoxin (TTX), Phentolamine and propranolol had no effect on the contractile effect of NT on air and ileum smooth muscle (P> 0.05) .The results suggested that NT may directly affect the NT-sensitive receptors on the empty and ileum smooth muscle membrane, and the jejunum smooth muscle The contractile enhancement after inhibition is related to opioid peptides and their receptors.
