目的:观察白细胞介素-4(IL-4)诱导人支气管上皮细胞(16HBE)不同时间血清类黏蛋白1样蛋白3(ORMDL3)的表达,探讨在细胞模型中IL-4对ORMDL3的转录和合成的影响。方法:用100ng/m L的IL-4诱导支气管上皮细胞后,分别在不同时间段(0、12、18、24、36、48h)用RT-PCR法检测ORMDL3 m RNA的表达,Western Blot法检测ORMDL3蛋白的表达。结果:正常支气管上皮细胞有微量的ORMDL3表达,IL-4诱导细胞后,ORMDL3m RNA和蛋白的表达均增加,在一定范围内呈时间依懒性,与对照组比较差异有统计学意义(P<0.05),其他各组与24h组比较差异有统计学意义(P<0.01)。结论:IL-4能诱导ORMDL3高表达,并且在一定范围内呈时间依赖性。
