目的研究2,3,7,8-四氯二苯并-p-二恶英(TCDD)在体外诱导肺癌细胞系SPC-A1细胞凋亡的作用。方法用MTT法测定TCDD对SPC-A1细胞的毒性,采用流式细胞仪对细胞凋亡进行分析,通过HE染色观察细胞形态变化。结果SPC-A1细胞生长曲线表明,随着TCDD浓度增高,SPC-A1细胞生长率明显下降;细胞凋亡可在TCDD浓度为10 nmol/L,0.1μmol/L,1μmol/L处理24 h后出现;TCDD诱导SPC-A1细胞的凋亡具有时间和浓度依赖性;凋亡细胞主要表现为细胞体积缩小,核染色质固缩,亚二倍体峰出现。结论TCDD在体外能有效诱导SPC-A1细胞发生凋亡。
Objective To investigate the effect of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) on the apoptosis of lung cancer cell line SPC-A1 in vitro. Methods The cytotoxicity of TCDD on SPC-A1 cells was determined by MTT assay. The apoptosis of SPC-A1 cells was analyzed by flow cytometry. The morphological changes were observed by HE staining. Results The growth curve of SPC-A1 cells showed that with the increase of TCDD concentration, the growth rate of SPC-A1 cells decreased obviously. The apoptosis of SPC-A1 cells appeared after treated with TCDD of 10 nmol / L, 0.1 μmol / L and 1 μmol / L for 24 h TCDD induced the apoptosis of SPC-A1 cells in a time-and-concentration-dependent manner. The apoptotic cells were mainly characterized by reduced cell volume, chromatin condensation and subduplex peaks. Conclusion TCDD can effectively induce apoptosis of SPC-A1 cells in vitro.
