目的建立反相高效液相色谱法测定斑蝥虫体内斑蝥素含量,用以监控斑蝥质量,指导临床用药。方法采用酸水加三氯甲烷从斑蝥虫体中提取斑蝥素的方法,然后进行含量测定,并与药典的提取方法的测定结果进行比较。结果用酸水加三氯甲烷提取斑蝥素测得的平均含量为2.840%,而用药典方法提取斑蝥素测得其平均含量为0.556%。用酸水加三氯甲烷提取斑蝥素测得的含量约是药典方法处理测得含量的5倍。结论酸水加三氯甲烷提取斑蝥素的方法简单方便,测得的斑蝥素含量比药典提取方法测定结果更能反映斑蝥虫体中斑蝥素的真实含量。
OBJECTIVE To establish an RP-HPLC method for the determination of cantharidin in cordyceps mites to monitor the quality of cantharidin and guide clinical application. Methods Acidic water and trichloromethane were used to extract cantharidin from plague worm body. Then the content of cantharidin was determined and compared with that of the pharmacopoeia extraction method. Results The average content of cantharidin extracted by acid water and trichloromethane was 2.840%, while the average content of cantharidin extracted by pharmacopoeia was 0.556%. Extract cantharidin with acid water and chloroform content of about 5 times the content of the pharmacopoeia. Conclusion The extraction of cantharidin from acid water and chloroform is simple and convenient. The measured content of cantharidin can better reflect the true content of cantharidin in the plague.
