高压电场对A549肺癌细胞株转移潜能影响研究

目的:研究不同强度的体外高压电场对肺癌细胞系A549细胞的黏附、迁移及侵袭等能力的影响。方法:选择处于生长周期的A549细胞,实验组共分为7个亚组,分别施加500~2 500V/cm的高压电场,对照组未施加电场处理。经生长抑制实验、黏附实验、侵袭及转移实验检验不同强度高压电场对A549细胞的影响。结果:1)细胞黏附能力:实验组各亚组的黏附细胞数明显少于对照组,实验组各亚组与对照组、实验组各亚组之间差异有统计学意义,P<0.001。2)细胞迁移能力:电场强度为500V/cm的实验亚组与对照组比较,差异无统计学意义,P=0.089;电场强度≥750V/cm的各实验亚组之间、及其与对照组比较,差异有统计学意义,P<0.001。3)细胞侵袭能力:电场强度为500和750V/cm的实验亚组与对照组比较,差异无统计学意义,P=0.181;电场强度≥1 000V/cm的实验亚组与对照组间比较,差异有统计学意义,P<0.001;电场强度为1 000~1 250V/cm的实验亚组和1 500~2 000V/cm实验亚组间比较,差异有统计学意义,P<0.001。结论:体外高压电场可以明显抑制A549细胞的黏附、迁移和侵袭等能力;随着电场强度的增加,肿瘤细胞的抑制程度也随之增加。 Objective: To investigate the effects of different intensities of high-voltage electric field on the adhesion, migration and invasion of lung cancer cell line A549. Methods: The A549 cells in the growth cycle were selected. The experimental group was divided into seven subgroups, and the high voltage electric field of 500-2 500V / cm was applied respectively. CONCLUSION: In vitro high-voltage electric field can obviously inhibit the adhesion, migration and invasion of A549 cells. With the increase of electric field intensity, the inhibition degree of tumor cells also increases.
