目的优选提取鲜马奶抗氧化活性肽的酶解条件。方法以水解度为指标,以酶解温度、酶解时间、加酶量为考察因素,筛选抗氧化活性肽提取工艺参数,测定其还原能力及对羟自由基清除、DPPH、超氧阴离子的清除能力。结果提取鲜马奶中活性肽的最佳酶解条件为温度45℃,酶解时间3h,加酶量3.5%。其还原能力为96.31%,DPPH清除能力为90.78%,超氧阴离子清除能力为94.28%,羟自由基清除能力为94.56%,与Vc相比无明显差异。结论鲜马奶中的活性肽具有良好的抗氧化活性,对DPPH、超氧阴离子、羟自由基均有较好的清除作用。优选得到的酶解工艺稳定、可行,可为抗氧化肽的研究提供理论依据和实验基础。
Objective To optimize the enzymatic hydrolysis conditions for the extraction of antioxidant peptides from fresh horse milk. Methods The degree of hydrolysis was taken as the index, and the factors of enzymolysis time, enzymolysis time and amount of enzyme were taken as the investigation factors. The technological parameters of antioxidative peptide extraction were screened, and the reduction ability and the removal of hydroxyl free radicals, DPPH and superoxide anion ability. Results The optimum enzymatic hydrolysis conditions for the extraction of active peptide from fresh horse milk were as follows: temperature 45 ℃, enzymolysis time 3h, enzyme dosage 3.5%. The scavenging capacity of DPPH was 96.31%, the scavenging capacity of DPPH was 90.78%, the scavenging capacity of superoxide anion was 94.28%, and the scavenging capacity of hydroxyl radical was 94.56%. There was no significant difference compared with Vc. Conclusion The active peptides in fresh horse milk have good antioxidative activity and good scavenging effects on DPPH, superoxide anion and hydroxyl radical. The optimal enzymatic hydrolysis process is stable and feasible, which can provide theoretical basis and experimental basis for the study of antioxidant peptides.
