目的探讨不同浓度促卵泡生成素(FSH)对卵母细胞体外成熟(in vitro matueation,IVM)的影响。方法选取8~12周龄雌鼠,腹腔注射HMG,48h后处死取卵巢,穿刺卵巢表面获得卵母细胞。将所得卵母细胞随机分组,置于不同浓度FSH(0,38,75,150,300IU/L)的培养基中,组别由A~E表示。培养24h后,脱去颗粒细胞观察卵子成熟率。对成熟卵行单精子显微注射,观察受精率及胚胎发育情况。结果1C、D、E组较其他组有较高的成熟率,差异有统计学意义(P<0.05),且这三组间差异无统计学意义(P>0.05)。2C、D组较其他组有较高的优质胚胎率(P<0.05),C、D二组间差异无统计学意义(P>0.05),但C组略高于D组。结论培养液中FSH浓度影响IVM结局:1当浓度<75IU/L时,成熟率下降。2当浓度在75~150IU/L时,胚胎发育较好。
Objective To investigate the effects of different concentrations of follicle stimulating hormone (FSH) on in vitro maturation (IVM) of oocytes. Methods Eight to 12-week-old female mice were injected intraperitoneally with HMG. After 48 hours, ovariectomized mice were sacrificed and oocytes were obtained by puncturing ovarian surface. The oocytes obtained were randomly divided into different concentrations of FSH (0, 38, 75, 150, 300 IU / L) medium, the group represented by A ~ E. After culturing for 24 hours, the granulosa cells were removed to observe the egg maturation rate. The mature egg microinjection of sperm injection to observe the fertilization rate and embryonic development. Results The rates of maturation in groups 1C, D and E were higher than those in other groups (P <0.05). There was no significant difference between the three groups (P> 0.05). There was no significant difference between groups C and D (P> 0.05), but the level of group C was slightly higher than that of group D (P <0.05). Conclusion The concentration of FSH in culture solution affects the IVM outcome: 1 When the concentration is less than 75 IU / L, the maturation rate decreases. 2 When the concentration of 75 ~ 150IU / L, embryonic development is better.
