目的检测转录因子维甲酸相关孤核受体α(retinoid acid receptor related orphan receptorα,RORα)、细胞表面受体T1/ST2和IL-17RB以及IL-5和IL-13等细胞因子的表达水平;了解胃癌患者外周血中nuocytes细胞的极化状态,探讨其与胃癌发生发展的关系。方法常规Ficoll-泛影葡胺密度梯度离心法分离外周血单个核细胞;实时荧光定量PCR检测30例胃癌患者PBMC中RORα、IL-17RB、T1/ST2、IL-5、IL-13 mRNA表达水平。结果胃癌患者外周血PBMC中RORα、IL-17RB、T1/ST2、IL-5 mRNA表达水平明显高于健康对照组,而IL-13的表达水平则明显低于健康对照组。相关性分析结果表明,胃癌患者RORαmRNA水平与IL-17RB、T1/ST2、IL-5、IL-13 mRNA水平呈正相关。结论胃癌患者机体存在着nuocytes极化状态,可能是构成胃癌患者Th1/Th2细胞平衡失调的基础,也与MDSC、M2巨噬细胞参与的免疫抑制微环境有着密切的关系。
Objective To investigate the expression of transcription factors such as retinoid acid receptor related orphan receptorα (RORα), cell surface receptors T1 / ST2 and IL-17RB, IL-5 and IL-13 and so on. The polarization state of nuocytes in peripheral blood of patients with gastric cancer and its relationship with the occurrence and development of gastric cancer. Methods Peripheral blood mononuclear cells were isolated by routine Ficoll-diatrizoate meglumine gradient centrifugation. The mRNA expression levels of RORα, IL-17RB, Tl / ST2, IL-5 and IL-13 in PBMC of 30 gastric cancer patients were detected by real- . Results The expression of RORα, IL-17RB, T1 / ST2 and IL-5 mRNA in peripheral blood of gastric cancer patients was significantly higher than that of healthy controls, while the expression of IL-13 was significantly lower than that of healthy controls. Correlation analysis showed that there was a positive correlation between RORαmRNA and IL-17RB, T1 / ST2, IL-5 and IL-13 mRNA levels in gastric cancer patients. Conclusion The existence of polarization state of nuocytes in gastric cancer patients may be the basis for the imbalance of Th1 / Th2 cells in patients with gastric cancer and the immunosuppressive microenvironment involving MDSC and M2 macrophages.
