目的:探讨间皮素(mesothelin,MSLN)siRNA慢病毒对人卵巢癌裸鼠腹腔移植瘤的治疗作用。方法:利用人卵巢癌SKOV3细胞建立卵巢癌裸鼠腹腔移植瘤模型。实验分为3组:治疗组、阴性对照组和空白对照组。治疗组应用携间皮素siRNA慢病毒液(LV-MSLN-shRNA)对裸鼠模型进行治疗,阴性对照组采用空载体(LV-MSLN-neg)慢病毒液进行治疗,空白对照组注射PBS治疗。从裸鼠生长一般情况、成瘤率、体质量差、瘤体质量和肿瘤转移部位数等方面观察治疗效果,Westernblotting法检测移植瘤组织中间皮素蛋白的表达情况。结果:用间皮素siRNA慢病毒液对荷瘤裸鼠进行注射的治疗组裸鼠的成瘤率、体质量差、瘤体质量及肿瘤形成部位数均明显低于空白对照组和空载体LV-MSLN-neg治疗组(P<0.01)。注射慢病毒液后瘤体组织中间皮素蛋白表达明显降低(P<0.05)。结论:间皮素siRNA慢病毒液对卵巢癌腹腔移植瘤的生长有显著抑制作用。
Objective: To investigate the therapeutic effect of mesothelin (siRNA) siRNA lentivirus on human ovarian cancer xenografts in nude mice. Methods: Human ovarian cancer SKOV3 cells were used to establish ovarian cancer xenografts model in nude mice. The experiment was divided into three groups: treatment group, negative control group and blank control group. The treatment group was treated with LV-MSLN-shRNA, the negative control group was treated with empty vector (LV-MSLN-neg) lentivirus, and the blank control group was injected with PBS . From the general growth of nude mice, tumor formation rate, poor body mass, tumor mass and the number of tumor metastasis sites to observe the therapeutic effect, Western blotting assay of transplanted tumor tissue mesothelin protein expression. Results: The tumor formation rate, body weight, tumor mass and the number of tumor-forming sites in nude mice injected with mesothelin siRNA lentivirus were significantly lower than those in blank control group and empty vector LV -MSLN-neg treatment group (P <0.01). The expression of mesothelin in the tumor tissue was significantly decreased after injection of lentivirus (P <0.05). Conclusion: The mesothelin siRNA lentivirus has a significant inhibitory effect on the growth of transplanted ovarian cancer.
