二草清肝汤对内毒素性肝损害小鼠防护作用的实验研究

目的:探讨二草清肝汤对内毒素性肝损害小鼠的防护作用及其机制。方法:将200只BABL/c小鼠按随机数字法均分成4组各50只,正常对照组、LPS组、二草清肝汤小剂量组、二草清肝汤大剂量组。二草清肝汤剂量按人与动物体表面积和体质量换算,计算出小剂量组为3 m L/kg,大剂量组为6 m L/kg,灌胃1次/d,共12 d;正常对照组和LPS组小鼠同时予同体积的0.9%氯化钠溶液灌胃。末次给药4 h后,除正常对照组外所有小鼠均腹腔注射LPS(10 mg/kg)。记录各组小鼠存活率,光镜和电镜观察组织病理学改变,全自动生化分析仪检测谷丙转氨酶水平,酶联免疫吸附法检测血清中肿瘤坏死因子-α和白细胞介素-10的浓度。结果:正常对照组、LPS组、二草清肝汤小剂量组、二草清肝汤大剂量组的存活率分别为100%、30.0%、75.0%、80.0%,与LPS组相比,正常对照组、二草清肝汤小剂量组、二草清肝汤大剂量组存活率明显升高(P<0.05),肝脏病理损害程度和死亡率减轻,ALT水平明显下降;比较各模型组小鼠血清中TNF-α和IL-10的结果(同时间点比较,二草清肝汤大、小剂量组TNF-α含量低于LPS组,IL-10则相反),二草清肝汤大、小剂量组组间比较,差异无统计学意义(P>0.05)。结论:预防性给予二草清肝汤能抑制内毒素性肝损伤小鼠肝脏炎症反应,明显减轻LPS所致的肝损害,其机制可能与降低TNF-α水平和增高IL-10水平有关。 Objective: To investigate the protective effect of Ercao Qinggan Decoction on endotoxin-induced liver damage in mice and its mechanism. Methods: 200 BABL / c mice were randomly divided into 4 groups of 50, normal control group, LPS group, Ercao Qinggan Decoction low dose group and Ercao Qinggan Decoction large dose group. The dosage of Ercao Qinggan Decoction was calculated according to body surface area and body weight of human and animal. The dose of small dose group was 3 m L / kg, the high dose group was 6 m L / kg, and gavage was given once daily for 12 days. Normal control group and LPS group mice were given the same volume of 0.9% sodium chloride solution gavage. Four hours after the last administration, all mice except the normal control group were injected intraperitoneally with LPS (10 mg / kg). The survival rate of mice in each group was recorded. The histopathological changes were observed under light microscope and electron microscope. The levels of alanine aminotransferase (ALT) were detected by automatic biochemical analyzer. The concentrations of TNF-α and IL-10 in serum were detected by enzyme-linked immunosorbent assay . Results: The survival rates of the low-dose group of Ercao Qinggan decoction of normal control group, LPS group and Ercao Qinggan decoction group were 100%, 30.0%, 75.0% and 80.0%, respectively. Compared with LPS group, The control group, Ercao Qinggan Decoction low dose group, Ercao Qinggan Decoction high-dose group was significantly higher survival (P <0.05), the degree of liver damage and mortality reduced, ALT levels were significantly decreased; The serum TNF-α and IL-10 results (at the same time point comparison, Ercao Qinggan Tang large and small dose group TNF-α content lower than the LPS group, IL-10 is the opposite), Ercao Qinggan Tang large , But there was no significant difference between low-dose groups (P> 0.05). CONCLUSION: Preventive administration of Ercao Qinggan Decoction can inhibit hepatic inflammatory response in mice with endotoxin-induced liver injury and significantly reduce hepatic injury induced by LPS. The mechanism may be related to decreasing TNF-α level and increasing IL-10 level.
