目的:研究扶正消癌合剂对人肺腺癌A549细胞(以下简称A549细胞)凋亡的影响。方法:制备大鼠含药血清,MTT法检测扶正消癌合剂含药血清对A549细胞增值的影响。Annexin V-FITC/PI双染流式细胞术观察含药血清对A549细胞凋亡的影响。Western Blot检测含药血清对P53,B淋巴细胞瘤-2基因(Bcl-2)及Survivin蛋白表达的影响。结果:扶正消癌合剂以时间、剂量依赖方式抑制A549细胞增殖,与DDP合用有协同增效作用,48、72、96h联合组的抑制率分别为28.27%、38.68%、37.98%;其可以诱导细胞的凋亡,且以晚期凋亡为主,凋亡率与对照组比较差异有统计学意义(P<0.05);其在诱导细胞凋亡过程中,下调Bcl-2、Survivin蛋白表达,同时活化P53蛋白表达。结论:扶正消癌合剂可能通过抑制Bcl-2、Survivin蛋白表达,同时活化P53蛋白表达诱导A549凋亡。
Objective: To study the effects of Fuzheng Xiaojiao Mixture on the apoptosis of human lung adenocarcinoma A549 cells (hereinafter referred to as A549 cells). Methods: Rat serum was prepared. The effect of Fuzheng Xiaojiao Mixture containing serum on the proliferation of A549 cells was detected by MTT assay. Annexin V-FITC / PI double staining flow cytometry of serum containing A549 cell apoptosis. Western Blot was used to detect the expression of P53, Bcl-2 and Survivin protein in serum containing serum. Results: Fuzheng Xiaojiao mixture inhibited the proliferation of A549 cells in a time-and dose-dependent manner and synergized with DDP. The inhibitory rates of Fuzheng Xiaojiao Mixture in combination group were 28.27%, 38.68% and 37.98%, respectively. (P <0.05). In the process of inducing cell apoptosis, the expression of Bcl-2 and Survivin protein were down-regulated while the apoptosis rate was higher than that of the control group Activated P53 protein expression. Conclusion: Fuzheng Xiaojian Mixture may induce apoptosis of A549 cells by inhibiting the expression of Bcl-2 and Survivin proteins and activating P53 protein expression.
