本研究以拟南芥AtMAPK3的核苷酸序列(Gen Bank accession No.NM_114433.2)作为查询序列进行BLAST搜索获得了马铃薯中的同源序列,将其命名为StMAPK3,其开放阅读框为1 122 bp,编码373个氨基酸。StMAPK3具有典型的促分裂原活化蛋白激酶的磷酸化位点TEY基序。从马铃薯栽培品种“大西洋”中克隆得到编码StMAPK3的基因序列,并构建了其植物表达载体p CPB-StMAPK3。用冻融法将重组质粒转入根癌农杆菌LBA4404中,然后再转化马铃薯栽培品种“大西洋”获得了转基因植株,为进一步明确MAPK在马铃薯信号转导过程中的作用提供帮助。
In this study, a BLAST search was performed using the nucleotide sequence of Arabidopsis AtMAPK3 (GenBank accession No. NM_114433.2) as the query sequence to obtain the homologous sequence in potato, which was named as StMAPK3 with an open reading frame of 1 122 bp, encoding 373 amino acids. StMAPK3 has a typical mitogen-activated protein kinase phosphorylation site TEY motif. The gene sequence encoding StMAPK3 was cloned from potato cultivar “Atlantic” and its plant expression vector pCPB-StMAPK3 was constructed. The recombinant plasmids were transformed into Agrobacterium tumefaciens LBA4404 by freeze-thaw method, and then transformed into potato cultivars “Atlantic ” to obtain transgenic plants for further clarifying the role of MAPK in potato signal transduction.
